## Introduction

Transmission of heritable information is the biological link between molecules, cells, organisms, and populations. DNA stores genetic instructions; chromosomes package that DNA; meiosis distributes chromosomes into gametes; fertilization restores diploidy; and evolutionary processes change allele frequencies across generations. On the MCAT, this topic often appears as a genetics passage involving pedigrees, crosses, linkage data, or population allele frequencies.

A strong approach is to connect levels of organization. A point mutation changes a DNA sequence. That change may alter a protein, phenotype, reproductive fitness, and eventually allele frequency in a population. MCAT questions often test whether you can move between these levels without confusing genotype, phenotype, gamete production, and population-level change.

### Meiosis, Fertilization, and Chromosome Behavior

Meiosis is the specialized cell division that produces haploid gametes from diploid germ cells. In humans, diploid somatic cells contain 46 chromosomes: 22 pairs of autosomes and one pair of sex chromosomes. Gametes contain 23 chromosomes, one from each homologous pair. Fertilization joins two haploid gametes to restore the diploid chromosome number in the zygote.

> A two-division process that reduces chromosome number from diploid to haploid and produces genetically variable gametes.

Meiosis has two major divisions. In meiosis I, homologous chromosomes separate. This is the reductional division because chromosome number is halved. In meiosis II, sister chromatids separate, similar to mitosis. A common exam error is thinking that sister chromatids separate in meiosis I. They do not; homologous pairs separate in meiosis I.

The key events that generate genetic variation are crossing over and independent assortment. Crossing over occurs during prophase I, when homologous chromosomes pair and exchange corresponding DNA segments. The visible connection points between homologs are called chiasmata. Independent assortment occurs at metaphase I, when homologous chromosome pairs line up randomly relative to each other. As a result, a gamete may receive different combinations of maternal and paternal chromosomes.

A useful estimate for independent assortment alone is:

where $$n$$ is the haploid number. In humans, $$n=23$$, so independent assortment can produce over eight million chromosome combinations, even before crossing over is counted.

Fertilization adds another source of variation because any sperm can combine with any egg. Together, recombination, independent assortment, and random fertilization explain why siblings, except identical twins, are genetically different.

Chromosome segregation errors can cause aneuploidy, an abnormal chromosome number. Nondisjunction is the failure of homologous chromosomes or sister chromatids to separate correctly. If nondisjunction occurs in meiosis I, all resulting gametes are abnormal because homologs failed to separate. If it occurs in meiosis II, some gametes may be normal and others abnormal. Trisomy 21, Down syndrome, is a classic example of aneuploidy caused by an extra chromosome 21.

> Do not treat mitosis and meiosis as interchangeable. Mitosis preserves chromosome number and produces genetically similar daughter cells. Meiosis halves chromosome number and produces genetically varied gametes.

### Mendelian Inheritance, Pedigrees, and Probability

Mendelian inheritance follows from chromosome behavior during meiosis. Mendel’s law of segregation states that the two alleles of a gene separate during gamete formation, so each gamete receives one allele. His law of independent assortment states that alleles of different genes assort independently if the genes are unlinked or far apart on the same chromosome.

For a single gene with two alleles, a heterozygote has two different alleles, such as Aa. A homozygote has two identical alleles, such as AA or aa. Dominant alleles affect the phenotype when one copy is present; recessive alleles usually require two copies to affect the phenotype.

If two heterozygotes for a fully dominant/recessive trait mate:

the expected genotype ratio is:

and the expected phenotype ratio is:

The MCAT often rewards probability logic more than memorized Punnett squares. The product rule applies when independent events must both occur. The sum rule applies when one of several mutually exclusive outcomes can occur. For example, if two carrier parents have an autosomal recessive condition in the family, the probability of an affected child is $$1/4$$ for each pregnancy. The probability of two affected children in a row is:

$$

Pedigrees require pattern recognition. Autosomal dominant traits often appear in every generation, affect males and females equally, and affected individuals usually have an affected parent. Autosomal recessive traits may skip generations; affected individuals can be born to unaffected carrier parents. X-linked recessive traits are more common in males because males have only one X chromosome. Affected males inherit the mutant X from their mothers and pass their Y chromosome to sons, so father-to-son transmission is not seen for X-linked traits. X-linked dominant traits can affect both sexes, but an affected father passes the trait to all daughters and no sons. Mitochondrial inheritance is maternal: affected mothers can pass the trait to all children, while affected fathers do not transmit mitochondrial DNA to offspring.

> **Test Tip:**
> In pedigree questions, first ask: does the trait skip generations? Are males and females affected equally? Is there father-to-son transmission? These three checks usually narrow the answer quickly.

Not all inheritance is simple Mendelian dominance. In incomplete dominance, the heterozygote has an intermediate phenotype. In codominance, both alleles are fully expressed; ABO blood type is the classic example, where IA and IB are codominant and both dominate i. Multiple alleles, penetrance, expressivity, pleiotropy, epistasis, and environmental effects can also modify phenotype.

Penetrance is the proportion of individuals with a genotype who show the phenotype. If a disease allele has 80% penetrance, not everyone carrying the disease genotype will show disease signs. Expressivity describes the severity or form of the phenotype among individuals who express it. These ideas matter in MCAT passages because a pedigree may not perfectly match simple ratios.

### Linkage, Recombination, and Genetic Mapping

Genes on different chromosomes assort independently. Genes on the same chromosome may be linked, meaning they tend to be inherited together because they are physically close. Crossing over can separate linked alleles, but the closer two genes are, the less likely a crossover will occur between them.

> The tendency of genes located near each other on the same chromosome to be inherited together.

Recombination frequency measures how often recombinant offspring appear in a genetic cross. A recombination frequency of 1% corresponds to 1 map unit, also called 1 centimorgan. If two genes have a recombination frequency of 8%, they are estimated to be 8 map units apart.

A recombination frequency near 50% indicates that two genes behave as if they assort independently. This can mean they are on different chromosomes or very far apart on the same chromosome. Recombination frequencies do not exceed 50% in standard mapping interpretation because 50% is the independent assortment limit.

> **Exam Warning:**
> A recombination frequency of 50% does not prove two genes are on different chromosomes. It means their inheritance pattern appears unlinked.

For linked genes, MCAT passages may describe parental and recombinant phenotypes. Parental types are the most common offspring classes because linked alleles are usually transmitted together. Recombinant classes are less common because they require crossing over between the loci.

Consider a testcross where an organism heterozygous for linked genes is crossed with a double recessive organism. If most offspring are AB/ab parental types and few are Ab/aB recombinant types, the genes are linked. The percentage of recombinant offspring gives the map distance. If 92 offspring are parental and 8 are recombinant out of 100 total, recombination frequency is 8%, so the genes are 8 map units apart.

Linkage can also explain why some dihybrid crosses do not produce the expected 9:3:3:1 phenotypic ratio. That ratio assumes independent assortment. If genes are linked, parental combinations appear more often than expected.

Sex linkage is a different idea from genetic linkage. Sex-linked genes are located on sex chromosomes, usually the X chromosome in MCAT genetics problems. Linked genes are genes physically near each other on any chromosome. A gene can be linked without being sex-linked.

> When a passage gives offspring counts, identify the largest classes first. They are usually parental types. The smallest classes are usually recombinants or double crossovers, depending on the mapping problem.

### Population Genetics and Evolutionary Forces

Inheritance explains how alleles move from parents to offspring. Population genetics explains how allele frequencies change across many generations. A population is a group of interbreeding individuals. A gene pool includes all alleles present in that population.

> A model describing allele and genotype frequencies in a non-evolving population under specific assumptions.

For a gene with two alleles, A and a:

p+q=1
$$

where $$p$$ is the frequency of allele A and $$q$$ is the frequency of allele a.

The genotype frequencies are:

$$

where $$p^2$$ is the frequency of AA, $$2pq$$ is the frequency of Aa, and $$q^2$$ is the frequency of aa.

Hardy-Weinberg equilibrium requires a large population, random mating, no mutation, no migration, and no natural selection. If these assumptions are met, allele frequencies remain constant from generation to generation. Real populations often violate these assumptions, so Hardy-Weinberg serves as a baseline model for detecting evolutionary change.

For example, suppose a recessive disease phenotype has frequency 0.04. For a recessive disease, affected individuals are aa, so:

q^2=0.04
$$

Therefore:

q=0.20
$$

and:

$$
p=1-q=0.80
$$

The carrier frequency is:

$$
2pq=2(0.80)(0.20)=0.32
$$

So 32% of the population are heterozygous carriers.

Genetic drift is not based on whether an allele improves survival or reproduction. It is random. Two special forms are the bottleneck effect and founder effect. A bottleneck occurs when a population is sharply reduced, leaving a smaller group whose allele frequencies may not represent the original population. A founder effect occurs when a small group starts a new population, carrying only a subset of the original genetic variation.

Gene flow, also called migration, occurs when individuals enter or leave a population and change allele frequencies. Mutation introduces new alleles. Nonrandom mating changes genotype frequencies and can increase homozygosity, though by itself it may not change allele frequencies unless paired with other forces.

> Differential survival or reproduction of individuals due to heritable traits, causing alleles associated with higher fitness to become more common.

Natural selection is not random with respect to fitness. If a trait increases reproductive success in a given environment, alleles contributing to that trait may increase in frequency. Selection can be directional, stabilizing, or major. Directional selection favors one extreme phenotype. Stabilizing selection favors intermediate phenotypes. major selection favors both extremes over the intermediate.

Fitness refers to reproductive success, not physical strength alone. A genotype with high fitness leaves more viable, fertile offspring. An allele can be harmful in one environment but beneficial in another. The sickle cell allele is a classic example: homozygosity can cause sickle cell disease, but heterozygosity can confer malaria resistance in certain regions, maintaining the allele through heterozygote advantage.

MCAT passages may link population genetics with molecular biology. A mutation in DNA can create a new allele. If the allele changes protein function and affects survival or reproduction, natural selection may alter its frequency. If the allele is neutral, drift may alter its frequency, especially in small populations.

> For Hardy-Weinberg problems, identify what the question gives you. If it gives the recessive phenotype frequency, that is usually $$q^2$$, not $$q$$. Take the square root before finding carriers.
